Pazopanib 400 mg Tablets

 

What is Pazopanib and what is used for :

Pazopanib is a is a medication that belongs to a class of medications known as” kinase inhibitors”. It is used to treat patients with the following indications:

Advanced renal cell carcinoma (advanced stage of kidney cancer). 

Advanced soft tissue sarcoma (advanced type of cancer that occurs in your soft tissue) in patients who have previously received chemotherapy.


Do not take Pazopanib :

If you are allergic to Pazopanib or any of the ingredients in this medicine. Tell your doctor about the allergy and what signs you had.


Warnings and precautions :

This medication may cause severe liver damage (may be fatal) and increased serum bilirubin and transaminase levels. Therefore, your doctor may recommend some liver blood tests prior to starting this medication and on a regular basis during the treatment. Your doctor may recommend dose adjustments accordingly. Make sure to tell your doctor if you have liver problems before starting this medication. Contact your doctor if you experience liver problem side effects such as pale skin, yellow eyes, white stools, dark urine, fatigue and swelling of abdomen.

This medication may cause QT prolongation and torsade de pointes (type of serious abnormal heart rhythm). Talk to your doctor if you experience signs like, lightheadedness, heart palpitations, weakness, blurred vision and seizures.  Make sure to tell your doctor about other medications that you are taking, or if you have low potassium levels in your blood, known QT prolongation or other heart problems in order to prevent QT prolongation from happening. Your doctor may recommend electrocardiograms and electrolytes monitoring depending on your case.

This medication may cause heart problems (e.g. heart failure which is a disorder that makes your heart muscle unable to pump blood properly). Your doctor may recommend monitoring your blood pressure and LVEF (ejection fraction; a test used to assess your heart’s ability to pump blood). Talk to your doctor if you have high blood pressure or heart problems before starting the medicine.  

This medication may cause fatal hemorrhagic events (bleeding events that can be deadly) and therefore should not be used in patients who have a history of hemoptysis (coughing up blood), or bleeding in the bowel, stomach or brain in the past 6 months. Make sure to tell your doctor before starting the medication if you have had similar bleeding problems in the last 6 months.

This medication may increase the risk of arterial thrombotic events (a blood clot that forms in your arteries) which can be fatal and therefore should be used with caution if you are at higher risk for similar events. Make sure to tell your doctor if you have previously experienced arterial thrombotic events (e.g. heart attack or stroke).

This medication may increase the risk of venous thrombotic events (blood clot that forms in your lungs or peripheries), including fatal pulmonary emboli (a blood clot that forms in your lungs). Contact your doctor immediately if you develop VTE signs and symptoms, like, shortness of breath, chest pain, or arm or leg swelling. Make sure to talk to your doctor before starting this medication if you have previously experienced a blood clot in your lungs or a vein. 

This medication may cause gastrointestinal perforation or fistula (tear or abnormal connection in your stomach or bowel) which can be fatal. Make sure to tell your doctor if you have a history of gastrointestinal perforation or fistula.

This medication may cause reversible posterior leukoencephalopathy syndrome (a rare brain-capillary leak syndrome). Stop taking the medication and contact your doctor immediately if you experience signs, like visual loss, seizures, headache and altered mental status.

This medication may cause hypertension (high blood pressure) and hypertensive crisis (severe increase in blood pressure that may lead to other complications) which are most likely to happen within the first few months of treatment. Your doctor may recommend monitoring your blood pressure before starting the medication, during the therapy, within one week after the treatment and regularly thereafter. Your doctor should make sure that your blood pressure is well controlled before starting you on this medication. Contact your doctor immediately if you experience high blood pressure signs like, headache, blurred vision, vomiting and severe anxiety. 

Make sure to tell your doctor if you are scheduled for a surgery or if you have undergone a surgery within the last 7 days. Your doctor should recommend stopping the medication 7 days before the planned surgery.   

This medication may cause hypothyroidism (underactive thyroid gland). Talk to your doctor if you have thyroid problems before starting this medicine. Your doctor may recommend monitoring for thyroid function tests. 

This medication may cause proteinuria (increased protein in your urine). Your doctor may recommend monitoring your urine protein and may adjust the dose or stop the medication depending on your urine protein levels.

This medication may increase the risk of serious infections which may be fatal. Talk to your doctor immediately if you experience signs of infection like, fever, sore throat, nasal stuffiness, pain or burning with urination, chills and sweats, cough, shortness of breath and stiff neck. 

This medication may cause harm to your baby if taken during pregnancy. Make sure to avoid pregnancy if you are taking this medication. 

Talk to your doctor before starting the medicine if you have a history of seizures, headaches or vision problems.  

Your healthcare provider will ask for urine, heart and blood tests before starting the medication and during the therapy.  


Possible side effects :

Possible side effects include signs of an allergic reaction, like rash and itching, high protein in your urine, signs of liver problems, like, nausea, vomiting, pain on the right side of your abdomen, yellowing of your skin or the whites of your eyes, tiredness, dark urine, loss of appetite and easy bruising, signs of heart problems, like fainting, fast or irregular heart rate, shortness of breath, leg edema, abdominal edema and fatigue, signs of heart attack or stroke, like difficulty breathing, chest pain, pain in your back, arms, jaw or neck, trouble talking, dizziness, headache and numbness or weakness on one side of your body, signs of blood clots (may be deadly) like, a pale or cool leg or arm, difficulty breathing, new chest pain, leg pain, and swelling of the legs, feet, arms and hands, signs of bleeding problems, like, bruising, unusual bleeding and delayed wound healing, signs of perforation or fistula in your bowel or stomach, like, vomiting blood, swelling in your abdomen, pain and black sticky stools, signs of reversible posterior leukoencephalopathy, like, high blood pressure, lack of energy, changes in vision or visual loss, seizures, confusion, headache and altered mental status, signs of high blood pressure, like severe chest pain or headache, loss of consciousness, confusion, blurred vision, nausea, vomiting, difficulty breathing, anxiety or seizures, signs of thyroid problems, like heat or cold intolerance, changes in heart rate, weight changes, hair loss and dry skin, signs of infection like, fever, wounds or scrapes (may be swollen, red, warm and painful), tiredness, nasal stuffiness, pain or burning with urination, body aches, chills and sweats, cold symptoms (e.g. cough, runny nose and sore throat), shortness of breath and stiff neck and signs of collapsed lung (air gets trapped in the space between the chest wall and the lung), like difficulty breathing. Most common side effects in advanced renal cell carcinoma patients include diarrhea, high blood pressure, nausea, hair color changes, anorexia and vomiting. Most common side effects in advanced soft tissue sarcoma patients include diarrhea, fatigue, bone and muscle pain, difficulty breathing, changes in skin color, hair color changes, change in taste sensation, hypertension, tumor pain, nausea, headache, decreased body weight, decreased appetite and vomiting.  


Other medicines and Pazopanib :

Tell your doctor or pharmacist if you are taking, have recently taken or might take any other medications including over the counter non-prescription medicines, prescription medicines, herbal supplements, vitamins and supplements.

If you are taking simvastatin (used to treat blood cholesterol).

If you are taking ketoconazole (used to treat fungal infections).

If you are taking medications used to treat human immunodeficiency virus (HIV) infection, like, ritonavir.

If you drink grapefruit juice.

If you are taking certain antibiotics (used to treat bacterial infections).

If you are taking medicines used to treat irregular heartbeats. 

If you are taking certain antidepressants (used to treat depression).

  

How to take Pazopanib :

Recommended dose is 800 mg orally once daily. Your doctor may recommend a reduction in your dose if you have liver problems: 200 mg orally once daily.  This medication is not recommended if you have severe liver damage.

Take tablet without food (at least 1 hour before or 2 hours after a meal).

Take tablet with a full glass of water. 

Do not crush the tablet. 

Grapefruit may increase the concentration of this medication in your body. Make sure not to eat or drink any grapefruit product while taking this medication. 

Your doctor will determine the specific dose that you need to take and may change the dose depending on your response. Take exactly as prescribed by your doctor. 


How to store Pazopanib :

Keep out of the reach of children and pets

Store medication at 20 to 25°C

Do not use beyond the expiry date mentioned on the box 

Keep medication in its original pack to protect it from moisture and light

Do not use damaged pills

Do not dispose medication in the wastewater or household waste
